  1. S-tag

S tag is the name of an oligopeptide derived from pancreatic ribonuclease A RNase A . If RNase A is digested with subtilisin , a single peptide bond is cleaved, but the resulting two products remain weakly bound to each other and the protein, called ribonuclease S, remains active although each of the two products alone shows no enzymatic activity. The N terminus of the original RNase A, also called S peptide, consists of 20 amino acid residues, of which only the first 15 are required for ribonuclease activity. This 15 amino acid s long peptide is called S15 or S tag . The amino acid sequence of the S tag is Lys Glu Thr Ala Ala Ala Lys Phe Glu Arg Gln His Met Asp Ser. It is believed that the peptide with its abudance of charged and polar residues could improve solubility of proteins it is attached to. Moreover, the peptide alone is thought not to fold into a distinct structure. On DNA level the S tag can be attached to the N or C terminus of any protein . After gene expression , such a tagged protein can be detected by comercially availabe antibodies . References R.T. Raines et al., The S Tag Fusion System for Protein Purification. Identifiers are relatively convenient for humans to mint create , read, type, remember etc. No central registration is necessary, at least for holders of domain names or email addresses and there is negligible cost to mint each new identifier. The identifiers are independent of any particular resolution scheme. Examples The following is an example tag that could be minted by a user to indicate authorship of a resource on the World Wide Web . The semantics of Tag URI do not require the user to currently have ownership of the Hostname upon which the tag is based, due to the date qualifier tag user example.com,2007 11 02 Tag URI The following example is a similar tag minted to indicate the name of the pet gerbil of the same user. 1. Myc-tag

A myc tag is a polypeptide protein tag derived from the c myc gene product that can be added to a protein using recombinant DNA technology. It can be used for affinity chromatography , then used to separate recombinant, overexpressed protein from wild type protein expressed by the host organism. It can also be used in the isolation of protein complexes with multiple subunits. A myc tag can be used in many different assays that require recognition by an antibody . If there is no antibody against the studied protein, adding a myc tag allows one to follow the protein with an antibody against the Myc epitope. Examples are cellular localization studies by immunofluorescence or detection by SDS PAGE protein electrophoresis. The peptide sequence of the myc tag is N EQKLISEEDL C 1202 Da . It can be fused to the C terminus and the N terminus of a protein. It is advisable not to fuse the tag directly behind the signal peptide of a secretory protein, since it can interfere with translocation into the secretory pathway .
