It is similar, conceptually, to two dimensional gel electrophoresis and difference gel electrophoresis in that it enables global identification of proteins that undergo altered electrophoresis electrophoretic migration resulting from, for example, proteolysis or post translational modification . For example, this approach can be used to compare samples from healthy and diseased patients. The methods for protein identification are identical to those used in general i.e. qualitative proteomics , but include quantification as an additional dimension. Mass spectrometry based proteomics Due to differences in ionization efficiency and or detectability the intensity of a peak in a mass spectrum is not a good indicator of the amount of the analyte. However, differences in peak intensity of the same analyte accurately reflect differences in its abundance. Therefore, in mass spectrometry based proteomics relative quantification is used to compare protein abundance between samples. This can be achieved by labeling one sample with stable isotope s alone or incorporated into protein crosslinkers like BisSulfosuccinimidyl suberate D4 BS3 , which leads to a mass shift in the mass spectrum. Differentially labeled samples are combined and analyzed together ... refbegin refend See also Protein mass spectrometry Quantitative proteomics bioinformatics stub Category Mass spectrometry Category Proteomics ...   more details

refimprove date March 2010 Image USAS12shotgun4104.jpg thumb 400px Daewoo USAS 12 automatic shotgun An automatic shotgun is a form of shotgun that is able to fire cartridges continuously while the trigger is pressed, unlike semi automatic weapons which require the user to depress the trigger for each projectile fired. Design Blowback firearms Blowback type weapons such as this use the force of the gas created by the accelerated deflagration of the propellant to cycle the action, ejecting the empty shell and loading another round without any user intervention. This process is similar to all automatic firearm automatic weapons , including machine pistol s and automatic rifle rifles . Selective fire shotguns provide an optional Semi automatic shotgun semi automatic mode through a switch. Almost all automatic shotguns are fed through some sort of Magazine firearms magazine , to provide the user with functions similar to an Assault rifle . 1. Shotgun messenger

In the late 19th and early 20th centuries, a shotgun messenger was a private express messenger and guard, especially on a stagecoach but also on a train , in charge of overseeing and guarding a valuable private shipment, such as particularly the contents of a strongbox on a stagecoach or safe on a train . The express messenger for stagecoaches typically rode in a seat on top of the coach, next to the driver. In the American Old West Old West of the 1880s, if a stagecoach had only a driver and no Wells Fargo messenger, this meant the coach carried no strongbox, and was thus a less interesting target for road agents bandits . Wells Fargo Co. express messengers typically carried a short or Sawed off shotgun sawed off 12 or 10 Gauge bore diameter gauge double barrelled shotgun , loaded with Lead shot buckshot . This was a most effective weapon in use against pursuing riders. Such weapons were sometimes referred to as messenger shotguns or, more commonly, coach gun s a name still used today . To some extent these weapons also carried over to use by private guards in trains with strongboxes or safes, where they were again effective.
