Autocrine signaling is a form of signaling in which a cell secretes a hormone or chemical messenger called the autocrine agent that binds to autocrine receptors on the same cell , leading to changes in the cell. ref Pandit, Nikita K. Introduction to the pharmaceutical sciences , page 238, 2007 ref This can be contrasted with paracrine signaling , intracrine signaling, or classical endocrine signaling. Examples An example of an autocrine agent is the cytokine interleukin 1 in monocytes . When interleukin 1 is produced in response to external stimuli, it can bind to cell surface receptor biochemistry receptors on the same cell that produced it. Citation needed date October 2009 Another example occurs in activated T cell lymphocytes , i.e., when a T cell is induced to mature by binding to a peptide MHC complex on a professional antigen presenting cell and by the B7 protein B7 CD28 costimulatory signal. Upon activation, low affinity Interleukin 2 IL 2 receptors are replaced by high affinity IL 2 receptors consisting of , , and chains. The cell then releases IL 2, which binds to its own new IL 2 receptors, causing self stimulation and ultimately a monoclonal population of T cells. These T cells can then go on to perform effector functions such as macrophage activation, B cell activation, and cell mediated cytoxicity . It binds to the protein CTLA 4 , which is expressed on the surface of activated T lymphocytes . Tremelimumab blocks the binding of the antigen presenting cell ligands B7.1 and B7.2 to CTLA 4, resulting in inhibition of B7 CTLA 4 mediated downregulation of T cell activation subsequently, B7.1 or B7.2 may interact with another T cell surface receptor protein, CD28 , resulting in a B7 CD28 mediated T cell activation unopposed by B7 CTLA 4 mediated inhibition. Cytotoxic T lymphocytes CTLs are generated by immune activation of cytotoxic T cells T sub c sub cells . They are generally CD8 sup sup , which makes them MHC class I restricted. CTLs are able to eliminate most cells in the body since most nucleated cells express class I MHC molecules. The CTL mediated immune system can be divided into two phases. In the first phase, functional effector CTLs are generated from naive T sub c sub cells through activation and differentiation. In the second phase, effector CTLs destroy target cells by recognizing the antigen MHC class I complex. Phase 1 In phase one, effector CTLs are generated from CTL precursors. The CTL precursors include naive T sub c sub cells since they are incapable of killing target cells. After a precursor cell has been activated, it can then differentiate into a functional CTL with cytotoxic activity. There are three sequential signals that are required to complete this process. First, there is TCR recognition of the peptide MHC class I complex. This step allows the cell to become licensed to an antigen presenting cell . Second, a costimulatory signal is transmitted by the interaction between CD28 and B7 of the precursor cell and the licensed antigen presenting cell. Last, a signal is induced by the interaction between IL 2 and the high affinity IL 2 receptor . This results in proliferation and differentiation of the antigen activated precursor cell into a functional effector CTL. Phase 2 In phase two, the now functional effector CTLs destroy the target cells. This can be done in two ways. These pathways are the cytotoxic protein pathway and the Fas ligand pathway. Apoptosis is the primary mechanism for both of these pathways. Cytotoxic Protein Pathway One pathway is the cytotoxic protein pathway. In this pathway perforins and granzymes are taken up by the target cell. In this pathway, the perforins facilitate the entry of granule contents into the cell. The granzymes then active the endogenous apoptosis pathw ... more details

PBB geneid 7293 CD134 , also known as OX40, is a member of the TNFR superfamily of receptors which is not constitutively expressed on resting na ve T cells , unlike CD28 . OX40 is a secondary costimulatory molecule , expressed after 24 to 72 hours following activation its ligand , OX40L , is also not expressed on resting antigen presenting cells, but is following their activation. Expression of OX40 is dependent on full activation of the T cell without CD28 , expression of OX40 is delayed and of fourfold lower levels. OX40 has no effect on the proliferative abilities of CD4 cells for the first three days, however after this time proliferation begins to slow and cells die at a greater rate, due to an inability to maintain a high level of PKB activity and expression of Bcl 2 , Bcl XL and survivin . OX40 binds to receptors on T cells, preventing them from dying and subsequently increasing cytokine production. OX40 has a critical role in the maintenance of an immune response beyond the first few days and onwards to a memory response due to its ability to enhance survival. OX40 also plays a crucial role in both Th1 cell Th1 and Th2 mediated reactions in vivo . OX40 binds TRAF2 , 3 and 5 as well as PI3K by an unknown mechanism. TRAF2 is required for survival via NF B and memory cell generation whereas TRAF5 seems to have a more negative or modulatory role, as knockouts have higher levels of cytokines and are more susceptible to Th2 meditated inflammation. TRAF3 may play a critical role in OX40 mediated signal transduction. CTLA 4 is down regulated following OX40 engagement in vivo and the OX40 specific TRAF3 DN defect was partially overcome by CTLA 4 blockade in vivo. TRAF3 may be linked to OX40 mediated memory T cell expansion and survival, and point to the down regulation of CTLA 4 as a possible control element to enhance early T cell expansion through OX40 signaling. OX40 has been implicated in the pathologic cytokine storm associated with certain viral infections, in ... more details
